Agamree Shows Promising Potential Beyond DMD Treatment: Catalyst Pharmaceuticals CPRX Reports Positive Phase I Data

Catalyst Pharmaceuticals (CPRX) has reported positive top-line results from a phase I study evaluating Agamree (vamorolone) for additional indications beyond its approved use in Duchenne muscular dystrophy (DMD). The study suggests that Agamree may deliver corticosteroid benefits without clinically meaningful immunosuppression, potentially opening the door to chronic inflammatory rare disease indications. The phase I study consisted of two parts involving healthy adult volunteers and assessed Agamree's glucocorticoid activity, anti-inflammatory effects, and immunosuppressive profile compared to Emflaza (deflazacort) from PTC Therapeutics (PTCT).

Santhera Pharmaceuticals (SIX: SANN) has announced top-line results from a Phase 1 clinical study of AGAMREE® (vamorolone) conducted by its North American licensing partner, Catalyst Pharmaceuticals, Inc. (CPRX). The two-part study evaluated AGAMREE® in healthy adult volunteers, assessing its glucocorticoid and anti-inflammatory activity while examining immunosuppressive potential compared to deflazacort.

In part A of the study, AGAMREE® and deflazacort demonstrated comparable cortisol suppression at clinical doses, with AGAMREE® showing less pronounced immunosuppressive biomarker effects. Part B revealed that clinically relevant immunosuppressive effects occurred only at the highest dose level, exceeding currently approved AGAMREE® dosing for Duchenne muscular dystrophy (DMD). These findings suggest that AGAMREE® may offer corticosteroid benefits with reduced immunosuppressive risks, potentially supporting its use in a broader range of chronic inflammatory rare diseases.

AGAMREE® is currently approved for DMD and is classified as a dissociative corticosteroid, designed to preserve anti-inflammatory efficacy while minimizing systemic side effects. Under the licensing agreement, Catalyst holds commercialization rights in North America, while Santhera retains right of first negotiation for new indications outside the region. The company is eligible to receive milestone payments and royalties based on net sales across all commercialized indications.
